Outline of Annual Research Achievements

1) レプチン応答性とTRIFシグナルとの関連について：（a）野生型マウスにTLR3リガンドであるpoly(I:C)を脳室内投与し，レプチン抵抗性が誘導されることを確認した。レプチン抵抗性の誘導がTRIF依存性かどうかについて、今後TRIF欠損マウスを用いて明らかにする。（b）TLR3欠損マウスに高脂肪食を摂餌させ、TRIF欠損マウスと同様の表現型を示すかどうか検討した。TLR3欠損マウスはTRIF欠損マウスとほぼ同等なやせ及び脂肪蓄積の減少を認めたが、摂餌量は野生型マウスと比べて変化を認めず、正常であった。
2) 視床下部の炎症病態とTRIFシグナルとの関連について：（a）野生型マウス及びTRIF欠損マウスに高脂肪食を摂餌させ、視床下部炎症におけるTRIFシグナルの関与について検討した。野生型マウスに比べてTRIF欠損マウスでは、高脂肪食による視床下部の炎症マーカー（TNF-α、MCP-1）の発現上昇が低下していた。またマクロファージマーカー（F4/80）の発現上昇も低下していた。（b）野生型マウスにpoly(I:C)を脳室内投与し、視床下部において炎症マーカー（TNF-α、MCP-1）の発現が上昇することを見出した。Poly(I:C)による視床下部炎症の誘導がTRIF依存性かどうかについて、今後TRIF欠損マウスを用いて明らかにする。
