Outline of Annual Research Achievements

細胞の皮膚と例えられる細胞膜は、厚さ6-10nmの脂質二重膜からなる自己組織化分子膜であり、酸素や水分子などは透過するが、電解質や高分子は透過できない半透膜としての性質を有する。細胞膜の破壊は、細胞内外でのイオン分配の解消や、それにともなう膜電位の脱分極、ATPなどエネルギー分子の細胞外漏出を引き起こし、細胞の持つ生命エネルギーを失わせる。そこで、本研究ではイオン応答性電界効果トランジスタ(ISFET)上に直接播種した細胞の脂質二重膜の状態をリアルタイム非侵襲に測定する方法を新たに開発した。本手法は、化合物の作用によって細胞膜上に形成された水素イオンなどの低分子量電解質が透過できる分子サイズの空孔を計測するという、これまでに無い新しい原理に基づく細胞膜障害性測定法である。従来法である赤血球溶血試験結果との比較を行ったところ、高い相関係数(r=0.88)が得られ、ISFET‐細胞によるリアルタイム・非標識な膜障害性の定量化が可能であることが明らかとなった。さらに、本システムではNH4+とH+などサイズの小さい(流体力学半径<0.33 nm)イオンの細胞膜透過を指標にしていることから、溶血試験におけるヘモグロビン(流体力学半径>3.1 nm)の透過が観察できないような微小な空孔形成をも検出することが可能であった。
近年の細胞工学では、遺伝子治療など細胞質に機能性分子を送達する需要が増しており、細胞膜透過ペプチドなどを始めとして、安全かつ高効率に細胞膜を透過するキャリア分子の開発が進められている。機能性キャリア分子の細胞膜透過機構を明らかにするためには高感度な細胞膜障害測定法の開発が必須の要件である。本研究の成果は先進治療にむけた細胞膜微小環境での生体分子動態の理解と、ナノ医療に伴う細胞毒性・治療効果の分子科学を構築し、将来的な医療イノベーションに向けた礎を構築するものである。
