研究概要

天然変性蛋白質の「折れたたみとカップルした結合」の機構を、計算機実験によって解明した。計算対象（天然変性蛋白質とパートナー分子、およびそれを取り囲む多数の溶媒分子）を全原子モデルで表現し、効率的構造探索法であるマルチカノニカル分子動力学を行った。それにより、折れたたみとカップルした結合過程の詳細な自由エネルギー地形を可視化した。実験的に決定された複合体構造以外にも、短い寿命の多様な複合体構造の存在を示した。得られた自由エネルギー地形をより大きな観点から理解するために、粗視化モデルでのサンプリングを行った。複数の準安定な複合体の間の構造遷移を、競合性と協調性の概念から表現した。
