研究成果の概要

東北大五味教授との共同研究として5種のベクターが使用可能な栄養要求性麹菌変異株を用いた異種遺伝子発現系を活用した糸状菌由来の生物活性物質の全生合成を検討した。その過程で、遺伝子導入を短縮し、かつ限られたベクター数の効率の良い使用が問題となったが、一挙に4個の遺伝子を導入する（Tandem法）を考案し、化合物種に関係なく遺伝子数4-17個を使って、構造の複雑な天然物が自在に合成できるシステムを開発した。このほか抗腫瘍性物質や抗生物質を含む放線菌由来の骨格構築酵素の機能解析や構造多様化に向けたハイブリッド酵素系の開発を検討した。その結果、生合成酵素の複合体の精妙な制御系を見出した。
