研究概要

ヒト免疫グロブリンH鎖遺伝子のエンハンサ-について,そのCisーacting elementsと解析し,さらにB細胞分化におけるエンハンサ-の役割について研究した。我々はヒトH鎖遺伝子エンハンサ-内に,二つのB細胞特異的転写に重要な役割を果たしているモチ-フを同定した。一つはヒトH鎖遺伝子JーCイントロエンハンサ-内の5'測に我々が見い出したHE2/μBエレメントであり,もう一つは3'側に新たに同定したE6エレメントである。以前に我々はHE2/μBエレメントに結合するB細胞特異的なDNA総合蛋白の存在を示したが,今回,HE2エレメント単独でエンハンサ-活性を示すこと,そのエンハンサ-活性は,B細胞内でのみ強く発揮されるが,T細胞はじめ他のいかなる非B細胞においてもエンハンサ-活性を示さなかった。この事から,H鎖遺伝子エンハンサ-のB細胞特異的転写活性化機能はHE2/μBエレメントの存在によると考えられた。さらにHE2エレメントをエンハンサ-として用いたSV40T抗原遺伝子のトランスジェニックマウスを作製したところ,H鎖遺伝子エンハンサ-全体を用いた場合と同様にB細胞系腫瘍がトランスジェニックマウス全部に発生した。このことから,HE2/μBエレメントはin vivoにおいてもB細胞特異的転写を誘導するエンハンサ-エレメントである事が示された。HE2/μBエレメントに結合するDNA結合蛋白を現在,サウスウエスタン法および機能的なアッセイ法によってcDNAクロ-ニングを行っている。今後、HE2/μBエレメントおよびその総合蛋白遺伝子について,ジ-ンタ-ゲディング法を用いて,B細胞分化における討割を検討してゆく。
