研究概要

アゾール系抗真菌剤は、真菌のP450(14DM)を阻害してその効力を発揮する.したがって,アゾール系抗真菌剤は,動物のP450に影響を与えずに真菌のP450(14DM)を阻害するという選択性を持つことが望ましい.本研究は,アゾール系抗真菌剤に選択性を与えるために有効な構造を推定するべく,P450(14DM)の基質認識に関係する構造を解析することを目的としたものであり,その成果は以下に要約できる.
(1)真菌のP450(14DM)は,基質であるラノステロールの3β水酸基,Δ8ステロールの骨格構造,およびステロール側鎖末端部分に対して構造特異的な認識を示す.(2)基質は,ステロール環をピロールCとヘム鉄の上におき,側鎖をピロールAに近接して存在するアミノ酸側鎖上に乗りあげさせた形で真菌P450(14DM)の活性中心に結合する.(3)ラットのP450(14DM)は,真菌のP450(14DM)と基本的に同じ様式で基質と結合すると考えられるが,側鎖末端部分に対する認識において明瞭な差違を示した.以上の知見は,P450(14DM)のステロール側鎖末端認識部位と相互作用するアゾール化合物は,真菌のP450(14DM)に対して選択性を示す可能性を示唆する.この可能性を確認するため,ステロール側鎖に相当するゲラニル基を有するアゾール化合物とその誘導体の酵母とラットのP450(14DM)に対する阻害作用を比較し,以下の知見を得た.(4)ゲラニル基を有する化合物は予期したとおり,側鎖認識部位に結合し,強い阻害作用を発揮した.(5)ゲラニル基を長さの異なるイソプレノイド基に置換した場合に見られる阻害力の低下は,酵母よりラットのP450(14DM)の方が顕著であった.(6)ラットの7-エトキシクマリン脱エチル化P450に対する阻害作用には,上記誘導体間での差があまり認められず,その阻害作用も弱いものであった.以上の知見は,P450(14DM)の側鎖認識部位と構造特異的に相互作用するアゾール化合物は,真菌の酵素に対する選択性を示す可能性を示唆する.
