研究概要

代謝型グルタミン酸、セロトニン2、TRH、ムスカリン3受容器応答はいずれもIP_3/Ca系という共通の2次メッセンジャー系を有するが、その詳細は必ずしも同一では無いこと、生体内ではこれらの情報はその入力順位が重要で、複数の情報入力に対して2次メッセンジャーレベルで情報の統合を行っていることが示唆された(赤池)。中枢神経細胞の興奮性は予想以上にグルコース代謝に依存し(赤須)、無酸素・無グルコース液中では5-6分で急峻な脱分極が生じる。これはNMDAとnon-NMDA受容器の活性と細胞内CaストアからのCa遊離により陽イオンの膜透過性が上昇することによる。また、低酸素により惹起される過分極はATP依存性Ca汲みだし機構の抑制により細胞内でのCa依存性Ca遊離が刺激され、カルモジュリンキナーゼIIとCキナーゼが活性化されるためと解った(東)。Ca依存性Ca遊離にはcyclicADP-riboseの合成が関与し、またミトコンドリアからのCa依存性Ca遊離機構が新たに発見された(久場)。細胞内Na濃度上昇が細胞内Ca濃度上昇を増強することも解った(反町)。シナプス前終末では上昇したCaと結合したカルモジュリンがミオシン軽鎖キナーゼを活性化して伝達物質の遊離に関与する(小林)。視床下部では体温調節中枢の温感受性ニューロンが高温域で内向き電流を惹起、それが“sleepy"チャネル様のNa電流であること、またPC-12細胞が温度感受性を持ち冷感受性ニューロンの特性解析のモデルになることも解った(清原)。そのほか神経系の初期発生過程(高橋)、膜電位依存性細胞内Ca遊離機構の獲得過程(井上)など進化・分化の側面、消化・吸収におけるイオンチャネルの役割(岡田、北里)、2次メッセンジャーによる軸索輸送の制御(竹中)、胃平滑筋の自動収縮の機構(富田)、血管平滑筋の内皮細胞依存性過分極(鈴木)、ヤリイカのClチャネル(山岸)、Naチャネル(瀬山、野間)、ギップ結合(外崎)についても検討し数々の新知見を得た。
