研究概要

マウスを用いて,食品抗原タンパク質の提示機構を解析した.B10.Aマウスを鶏卵白リゾチームで免疫した場合,87-96特異的T細胞の応答は誘起されないが,ペプチド85-96で免疫すると誘起され,T細胞抗原決定基87-96/Ekはクリプティック(潜在的)であった.またC3H/Heマウスに牛乳αs1-カゼインで経口寛容を誘導した場合,ドミナント(優勢)な抗原決定基に特異的なT細胞には寛容が誘導されるにもかかわらず,クリプティックまたはサブドミナント(劣勢)な抗原決定基に特異的なT細胞には寛容が誘導されないことが判明した.87-96/Ekに対する応答を詳細に調べた結果,1)抗原提示細胞(APC)によって提示する抗原決定基は異なること,2)in vivoにおいて87-96/Ekに応答するT細胞が活性化されない理由は,マウス体内でT細胞を初期に活性化するAPCは,in vitroの実験で観察された応答に関与するAPCとは異なり,87-96/Ekを提示できないためであることが示唆された.また,インターフェロンγは,抗原決定基の提示されやすさに影響を与え,その影響は抗原決定基ごとに,またAPCごとに異なっていた.以上のことより,抗原提示細胞によって特定の抗原決定基の提示されやすさは異なり,さらに炎症で分泌されるサイトカインも提示されやすさに影響を与えることが判明した.したがって,自己免疫疾患や食品アレルギーにつながる過剰な免疫応答には,何らかの要因によって抗原決定基の提示のされやすさが変化し,寛容になっていないT細胞の過剰な応答を誘導することが原因であるというモデルを立てることができた.このことは,アレルギー抑制法を開発するうえでも有用である.
