研究概要

平成5年度から8年度の4年間に亘って遂行された特別推進研究の代表的成果は以下のものである。
1)サイトカイン共通信号伝達分子gp130を介する信号を核内に伝達し、遺伝子発現を誘導する分子APRF/STAT3の遺伝子クローニングに成功し、その全構造を決定。その結果、gp130から遺伝子発現に至るシグナル伝達の2つの経路すなわち i) Ras-MAPカスケードからNF-IL6の活性化と ii) JAK-STATの全容を解明。
2)サイトカインのシグナル伝達に関わる分子の生体内における生理的、病理的役割を解明するため、gp130、NF-IL6をコードする遺伝子を破壊したノックアウトマウスを作成し、gp130は心筋細胞のアポトーシスの防御、増殖、分化に必須のシグナルを与えていることが明らかとなった。
3)転写因子NF-IL6ノックアウトマウスは細菌感染に対する防御、特にマクロファージが関与する防御が完全に失われることが明らかとなった。このマウスにおいてはγ-インターフェロンやNO(一酸化窒素)の生成は全く正常であるにも関らず、マクロファージは全くリステリアを殺せないことにより、NO非依存性の殺菌機構の存在を発見した。
4)ケモカイン(PBSF/SDF-1)のレセプターをクローニングし、これがHIVのcoreceptor (fusin)であることを発見した。
5)CD40ノックアウトマウスを作成し、このマウスがヒトの高IgM免疫不全症の患者と全く同じ症状を呈することにより、CD40-CD40Lの異常がこの病気をひきおこすことが確認された。
6)TGF-β、 PDGF-B、 IL6-IL6R遺伝子をラット肺上皮細胞に移入し、発現させ、種々の間質性肺炎(IPF、 ILP等)の発症誘導に成功。
7)ヒト型化抗IL-6R抗体を用いたミエローマ、キャッスルマン、リウマチ様関節炎の治療実験を開始し、その有効性を確認した。
