研究概要

ミトコンドリアタンパク質のミトコンドリアへの取り込みには,ミトコンドリアの外(サイトゾル)と中(マトリクス)の分子シャペロン,hsp70が関与している。本研究により,ミトコンドリアタンパク質前駆体には,折れたたみの過程で,プレ配列部分と成熟体部分が相互作用することによって凝集する危険性があること,サイトゾルのhsp70は両親媒性のプレ配列に結合することが明らかになった。したがってhsp70は,前駆体の成熟体部分に結合するだけでなく,前駆体の生合成時にまずアミノ末端のプレ配列部分に結合し,そのことによって前駆体の凝集を防ぎ,前駆体をミトコンドリアへの取り込み可能な状態に保つというシナリオが考えられる。
大腸菌において,Dnak(hsp70)は,DnaJ(hsp40)およびGrpEと協同して働くと考えられている。われわれは,酵母のGrpEホモログであるYgelp(Mgelp)がミトコンドリアのマトリクスに可溶性タンパク質として存在し,マトリクスのhsp70と協同して,ミトコンドリアタンパク質の膜透過を助けていることを見いだした。したがって,大腸菌において,Dnak,DnaJ,GrpEという三つのタンパク質が協同して働いていたのと同様に,真核生物のミトコンドリアにおいても,hsp70はパートナータンパク質であるDnaJホモログ,GroEホモログと協同して働いていることが,強く示唆された。
