研究概要

家畜を対象とした胚操作は、最終的には、子宮内への胚移植、着床・胎盤形成を経て、出産され、個体として成長して完結する。一連の過程は、さまざまな、生殖工学ないしは、発生工学の技術を必要とするが、特に、胚移植後、着床・胎盤形成過程における胚の損失が多く、移植胚の着床効率を上げることが、獣医・畜産学のみならず、医学においても重要な課題となっている。
本研究の主たる目的は、イスラエル側のマクロファージの分子・細胞生物学的研究に関する知識・技術や、家畜のトランスジェニック作出に関する知識・技術の提供を受けると同時に、わが国側で、東大牧場のクローズドコロニーとして維持されているシバヤギを材料として、子宮上皮細胞、トロフォブラスト細胞、ならびに、マクロファージのそれぞれ培養系を確立し、モデル系を用いて、in vitroで、着床現象におけるマクロファージの役割を解明し、胚移植後の着床効率を高めるための基礎知識を得ることにある。
この目的を達成するために、Ahuva Knyszynski博士の来日を仰ぎ、シバヤギの培養細胞系確立を試みた。昨年度から、東大牧場のシバヤギの感染症による幼弱期における死亡率が高くなり、材料の確保が困難になるという不測の事態が生じ、研究の進展も思うにまかせぬこととなったが、子宮上皮細胞系については、まだ、クローン化されてはいないが、培養細胞株を確立することができた。現在、クローン化の実験を進めている。この細胞系は、in vitroで管腔形成や、分泌顆粒を豊富に持った細胞への分化など、興味深い挙動を示し、クローン化が終了し次第、解析的実験を行う計画である。一方、トロフォブラスト細胞については、予備的にトロフォブラスト細胞と判定された細胞について、培養系の確立を図り、明らかに繊維芽細胞とは挙動の異なる培養細胞系を得たが、実験に使用する目的で継代・増殖を図る過程で、増殖率が急激に低下し、現在、その原因と対策を模索している。増殖率が回復し次第、細胞表層マーカーに対する抗体によるアフィニティーコラム法で、繊維芽細胞など、混在する非トロフォブラスト性細胞の除去、適当なマーカーを用いたトロフォブラスト細胞の同定などを行う予定である。マクロファージについては、マウスやヒトで主に用いられている方法を用いて、シバヤギからマクロファージの培養系を得ることを試みたが、残念ながら、2年間の 研究期間中に、適当な系を得ることはできなかった。シバヤギマクロファージのin vitroにおける挙動が、マウスやヒトのそれと異なる理由は明確でなく、今後、継続した研究が必要である。なお、得られたマクロファージを、レトロビールスを用いて不死化する手法も知られているが、われわれの研究室では、バイオハザード管理の適当な実験室が無く、シバヤギマクロファージ不死化の実験は、得られたマクロファージをイスラエル側に送って実験する計画であったが、マクロファージの適当な採取・培養法の確立ができぬ段階で、研究が終了することとなった。なお、イスラエル側では、マクロファージの活性化におけるシグナルトランスダクションについて研究を進め、主として、ラットおよびマウスのマクロファージを用いて実験を行い、MAPキナーゼの関与について、興味深い知見が得られつつある。なお、イスラエルでは、ヒト血液から分離したマクロファージを、床ずれ患者にIV注射で投与し、治療に用いる実験が、かつて、Weizmann研究所の超微形態部門の教授であり、Ahuva Knyszynski博士の共同研究者であったD.Danon教授を中心に、すでに臨床レベルで開始されている。その、最新の成果についての未発表情報を、今回の研究を通じて得ることができたことは、今後マクロファージに関する知識を、着床率の改善へ応用することを考える上で有益であった。また、着床における、CSF-1やインターロイキンファミリー、TNF-αをはじめとするサイトカインの役割の研究で著名な、B.Tartakovski博士は、従来Weizmann研究所で研究を行っていたが、現在はWeizmann研究所を離れ、企業で研究を続けている。今回の研究期間中、B.Tartakovski博士と、着床におけるマクロファージや他の免疫細胞の役割解明と、自己免疫疾患治療法の開発に関する意見の交換を行うことができた。
一方、われわれの研究室では、マウスならびにシバヤギを用いて、胎盤におけるホメオボックス遺伝子群の発現動態の解析、マトリックスメタロプロテアーゼcDNAに関する研究を行い、興味深い知見をえることができた。これらの知見は、今後、トロフォブラスト細胞や子宮内膜細胞の動態調節における、LIFをはじめとするサイトカインの役割を解明する上で、重要な手がかりになるものと思われる。今回の研究期間中に、実際、LIF(マクロファージが分泌することが知られている)がマウストロフォブラスト細胞において、膜型MMPの発現を促進するらしい事実が明らかになった。
さらに、マウスを用いた卵成熟の研究で著名な、Nava Dekel博士に来日を依頼し、家畜における胚操作の一環として重要な、in vitro卵成熟におけるシグナルトランスダクション系の役割に関する最新の研究成果や技術の情報を得ると共に、Weizmann研究所で所有するMRI装置によるラット着床部位や時期の非破壊的検出と、その着床・胎盤形成研究における応用について、今後の共同研究の可能性を論議した。また、イスラエルでトランスジェニックヤギの作出に成功しているMoshe Shani博士には、トランスジェニックヤギ作出の実際の作業を見学させてもらうことができた。また、博士に来日を依頼して、博士の研究グループの最新の研究情報を取得するとともに、今後の相互の研究交流の端緒を開くことができた。
今回の共同研究は、東大牧場のシバヤギの繁殖率の低下、Weizmann研究所における研究体制の大幅な改組など、不測の事態が生じ、当初予定した研究計画を消化することが困難であったが、今回の研究を端緒として、現在も、両サイドで研究を継続しており、今後、さらに、重要な成果が、日本、イスラエルの双方で挙げられることが期待される。
