研究概要

RecA蛋白質は、原核生物の遺伝的組換えにおいて中心的役割を果たす蛋白質である。近年、多くの真核生物においてもRAD51などのRecAホモログが見つかってきている。これらRecA様組換え蛋白質は、遺伝子組換えの他、遺伝子の発現調節、DNAの複製にも大きく関わるが、その機能発現機構には未解決の現象がまだ多く残されている。一般に、耐熱性菌の蛋白質は非常に安定性が高く、X先結晶解析をはじめとする物理化学的な測定に適していることが知られている。そこで本研究では、進化的に大きく異なる古畑菌・原核生物・真核生物のRecA蛋白質の比較解析から、その機能発現機構を解明することを目的として、まず耐熱性菌・古細菌のrecA遺伝子のクローニングを試みた。
まず、ロシア国内の温泉から、Methanosarcina sp.TS-2、Halococcus turomenicus B1734、Desulfurococcus amynoliticus、Halobacterium distributum B1733、B1739などの多くの熱耐性菌を採集した。RecA?RAD51で高度に保存されているアミノ酸配列を元に作製したプライマーを用いて、ゲノムからPCR法によってDNA断片を増幅し、これをプローブとしてゲノムDNAの遺伝子バンクから、recA/RAD51蛋白質と比較することにより、すべての生物種に共通で、しかも機能発現に必要な領域を明らかにした。また、この実験の過程で、D.amynoliticusゲノムDNAからトポイソメラーゼと有意な相同性を示す新しい遺伝子も見出された。この遺伝子産物の機能は今のところ不明であるが、古細胞における組換え機能を考える上で非常に興味深いものであり、これについても現在解析中である。
次に、全ての生物種に共通する組換え機構の基本的性質を明らかにするために、遺伝子操作系が確立した生物の中で、最も高温で棲息する代表的な耐熱性原核生物の高度好熱菌Thermus thermophilus HB8からrecA遺伝子をクローニングした。遺伝子操作を利用して量産化後、精製した好熱菌RecA蛋白質は、DNA鎖交換反応、DNA依存性ATP加水分解などの活性を有しており、これらが相同的組換え反応に必須の性質であることが示された。好熱菌RecA蛋白質の最大活性は70℃付近にあり、常温菌酵素と比べると、熱や変性剤に対してはるかに安定であった。この安定性を利用して、X線結晶構造解析のための結晶化を行ったところ、容易に結晶を得ることができた。また、常温菌酵素では困難であったDNA依存下での結晶化にも成功した。これらについては、現在解析データを収集中である。さらに,好熱菌RecA蛋白質は一部のドメインを取り除いても安定な構造を保持できることから、NMRによる立体構造解析のために、分子同士の会合ができない欠失変異体を作製した。これによって、従来のように重合することなく、高蛋白質濃度でのNMR測定が可能になった。また、会合を抑えるために構造を破壊することのない低濃度の界面活性剤の使用も問題なく行うことができた。今後、これらの構造解析研究によって、RecA蛋白質の機能発現に必要な立体構造および反応に伴う構造変化が明らかになるものと期待できる。
真核生物の組換えについては、植物のユリの2つのRecA蛋白質のホモログRad51およびLim15の減数分裂期染色体における分布を間接蛍光抗体法によって観察した。その結果、これらの蛋白質は、原核生物のRecA蛋白質と同じく、組換え反応の初期過程であるDNAのホモロジーサーチから染色体対合において、協同して重要な役割をしていることが明らかとなった。さらに、出芽酵母の減数分裂期に働くMRE2遺伝子を同定し、変異体などの解析から、pre-mRNAのスプライシングを介して交叉型組換え体の形成に関与していることが示唆された。
