研究概要

海馬培養神経細胞に対してglucocorticoidが神経細胞死を促進し、estrogenが10^<-6>〜10^<-9>の範囲では、これと拮抗的にneuriprotectiveに働くという我々の実験結果を前提として、神経細胞死に対するestrogenの作用の分子生物学的メカニズムを探るのが、本研究の目的である。
estrogenがin vivoでneuroprotectiveな作用を発揮するためには、当然その受容体が存在しなければならない。estrogen受容体は、周生期には、大脳、小脳を始め脳内に広く分布し、神経突起の伸長、神経回路網の形成に役割を演じた後、急激に消衰して、その分布が視床下部、辺縁系に限局することが知られている。in vivoでの神経細胞傷害後、estrogen受容体が出現するか否かが問題となる。
以上の考えに基づいて、実験をすすめ、次のような結果を得た。
1.estrogenが10^<-6>〜10^<-9>の範囲内ではneuronに対してprotectiveに働き、10^<-5>以上の濃度ではneurotoxicに働くという我々の実験結果をふまえ、estrogenのneuroprotectiveな作用のメカニズムを探るための実験が行われた。hepatocyte growth factor(HGF)と並んでinsulin-like growth factor(IGF)がこのメカニズムと関わっている可能性があり、従来から行ってきたLDH effluxmetryの手法を用いてIGFのNeuron survivalに対する影響を観察した。その結果、IGFIおよびII、ともにラット辺縁系培養神経細胞のneuron survival rateを増加させることを認めた。
2.estrogenの神経可塑性機能の発現の前提条件としてラットにカイニン酸を投与し神経細胞に傷害を与えた後、周生期と同じように大脳にestrogen受容体が再出現するか否かをestrogen受容体に対するモノクローナル抗体を用いて免疫組織化学的に検証した。その結果、ラットの大脳皮質の広い領域にわたって、ことに前頭部に著明にカイニン酸による神経細胞の傷害に引き続いてestrogen受容体が発現することを認めた。
3.カイニン酸投与によって、estrogen受容体が大脳皮質に再出現し、辺縁系ではその増加を認めるという事実を,in situ hybridizationによって、estrogen受容体の体場からも確認した。ことに、海馬CA3領域において著明であった。さらに、脳成熟後にestrogen受容体が存在する辺縁系では、その明らかな増加を認めた。
4.カイニン酸全身投与に先立ってestrogenを投与することにより、HGF、IGFI及びIIなどの神経栄養因子mRNA発現が、estrogenを投与しない場合と比べてどの様に影響されるか観察した。その結果、カイニン酸注射後3日後に観察した限りでは、estrogen投与によってIGF ImRNA発現の著明な上昇、IGF IIの同様な傾向がdot blot法によってみられた。
5.カイニン酸全身投与に先立ってestrogenを投与することにより、HGF、IGFI及びIIなどの神経栄養因子mRNA発現の上昇が見られることを認めたので、そのsignal transductionする経路を追求することとし、その第1段階として、estrogen単独投与によるAP-1結合活性の時間的推移をgel shift assayによって観察した。estrogen投与後、30〜120分にかけてAP-1結合活性の持続的上昇が見られることを認めた。
6.カイニン酸全身投与に先立ってestrogenを投与することにより、IGF I mRNAの上昇が見られることをdot blot分析法で認めたので、この点をさらにin situ hybridizationによって確認するとともに免疫組織化学染色によってIGFそのものの上昇が起こることを認めた。
7.カイニン酸全身投与に先立ってestrogenを投与することにより、ラット海馬を中心としてGAP43の出現が見られることを免疫組織化学的に観察した。
8.以上の結果から、estrogenがカイニン酸による神経細胞死に対してestrogen受容体の増加、AP-1結合活性の上昇、IGFIの誘導、GAP43の出現という経路をとって、神経修復に関与している可能性があるものと考えた。
