研究概要

本研究の目的は、過剰な塩・乾燥ストレスのシグナルを植物がどのように受容し、それらに応答し、さらに馴化していくかというメカニズムを解明し、それらを応用して過酷な環境条件、特に塩過剰環境・乾燥環境に耐えうる新しい植物の作出の可能性を探ろうとするものであった。中でも、1)プロリン、ベタインおよびポリオール等の適合溶質の生成機構、2)塩の植物体内で処理機構、3)種々遺伝子の植物への導入の方策とそれらを評価する方法を探ることであった。3年間に大きな前進を見た。それはプロリンやベタインの合成系をほとんど解明できたこと、またそれらに関係する遺伝子を植物体内に導入し、プロリンやベタインを大量発現する変異植物を作り、どれほど塩・乾燥環境に耐えるようになるかを評価したことである。その結果、これらの変異植物の塩・乾燥に対する耐性は、かなりのものであったが、既に知られている天然の耐塩・耐乾燥植物のそれには及ばなかった。これらの結果を受けて耐塩・耐乾燥性の獲得のために更なる遺伝子解析を行う必要が生じた。また、篠崎らは、外部環境からのシグナルがどのように伝達され、プロリン合成やベタイン合成遺伝子に作用するかというシグナル伝達機構を解明し、直接合成に関わる遺伝子の導入はもちろんであるが、シグナル伝達に関わる遺伝子を導入することによって、一層効率の良い遺伝子導入変異植物作出の可能性を提唱した。植物ゲノムの全遺伝子解析が進む中で、本研究の目的にもDNAマイクロアレーの利用が有効であるとの提案がボーナート教授によってなされた。このテクニクは発現遺伝子の探索やノーザンブロットなど多くの利用が考えられ、今後の植物のストレス応答の研究の発展にも重要であろう。
