研究概要

1. ウシ・フィラリア(Seteria digitata)をスリランカにおいて採取し、種々のクロマトグラフィー法を併用して、2種のペプスタチン感受性アスパラギン酸プロアアーゼを完全精製し、また数種の中性プロテアーゼを部分精製した。これらの標品について種々の分子的、酸素的性状を検討し、部分アミノ酸配列の解析を行った。
2. ヒト・フィラリアプロテアーゼのcDNAライブラリーをBrugia malayi(成虫および幼虫)、Onchocerca volvulus(成虫)、Brugia pahangi(幼虫)について調整した。また、遺伝子ライブラリーをBrugia malayiおよびWuchereria banerofti(幼虫)について調整した。また、Schistosoma mansoniカテプシンDのcDNAを増幅し、これを用いて、Brugic malayiより2種のアスパラギン酸プロテアーゼの全長cDNAと1種のcDNAを単離し、それらの塩基配列を決定し、一次構造を推定した。このうち、2種はカテプシンD様プロテアーゼであり、1種はカテプシンD、ペプシンのいずれともかなり異なるユニークなグループに属することが示唆された。
3. Brugiamalayi cDNAライブラリーより2種のシステインプロテアーゼの全長cDNAを得、それらの塩基配列を決定して、一次構造を推定した。パパイン様酵素と考えられるが、各種カテプシン系シツテインプロテアーゼのどのグループに属するかは明瞭ではない。
4. 上記研究を並行して、その基礎となる研究と各種のプロテアーゼとプロテアーゼイインヒビターについて行った。プロガストリンC末端切断プロテアーゼ(ブタ胞)、Rasタンパク質C末端CAAXモチーブ特異的プロテアーゼ(ウシ胞)、エンテロペプチダーゼ(ブタ十二指腸)、トリプシン及びキモトリプシン、ミクロソーム膜結合セリンプロテアーゼ(ラット肝)、コロプシン(ブタ中腸)、カテプシンE(マウス、ヒト、胃)、シーラカンスベプシノーゲン、その他約10種のプロテアーゼについて、それらの精製分子的・酵素的性状解析、構造・機能機関の解析等を行い、またプロテアーゼインヒビター(プロメラインインヒビター、ペプシニンヒビターなど)についても同様な研究を行った。
