研究概要

本研究の目的は,1)ヒト肝癌発症の原因ウイルスと考えられるB型およびC型肝炎ウイルスの機能的受容体の構造と性質を解明し、ウイルス性肝癌の発症機構における宿主受容体の役割を明らかし,2)これにより肝癌発症の予防、診断、治療への基礎的情報を得ることである。今年度の研究において,上記の目的をほぼ達成できた.すなわち,
(1)ヒト肝臓からB型肝炎ウイルス表面抗原(HBsAg)と特異的に結合する糖タンパク質の部分精製に成功し,これに対するモノクローナル抗体の作成も達成した。
(2)ウイルス表面抗原(HBsAg)に対する受容体は,ヒト肝臓においてはシヌソイドに局在することを見出した.また,この結合にはシアル酸が関与することが示唆された.
(3)この糖タンパク質は,Neu5Acα 2-6Gal、Neu5Acα 2-3Galおよび、Galβ1-3GalNAcを含有するムチン型糖鎖からなることを見い出した。
(4)ヒト膵癌細胞から得たムチン画分にもウイルス表面抗原(HBsAg)が結合することを見出し,B型肝炎ウイルス受容体物質の構造解析に有用な系口が得られた.
(5)C型肝炎ウイルスの膜表面抗原を発現する細胞からこれらを特異的かつ効果的に遊離させる界面活性剤を見出した.これによりC型肝炎の受容体をアッセイできる可能性が開けた.
以上,B型肝炎の受容体についてはかなり分子レベルの情報が得られ,ヒト肝臓からウイルス抗原と結合できる糖タンパク質を実際に分離することが出来,その組織特異性も明らかに出来た.今後,この糖タンパク質の完全精製,遺伝子のクローニング,発現系の構築などにより,感染制御に結びつく成果が期待される.また,C型肝炎ウイルス受容体についても,今まで不可能であった受容体アッセイ系が可能となり,ウイルス膜抗原の十分な量が確保出来れば,受容体分離も可能であると思われる.
