研究概要

カイコの胚発生および後発生における遺伝子発現様式を遺伝学,形態学,生化学,分子生物学的手法で追求して総合的に考察した.まず,ハウスキーピング遺伝子の典型としてアルドラーゼアイソザイムを粗識別に検定し,精製して酵素的性質とアミノ酸配列を解析する一方,cDNAを単離しそれをプローブとして器官特異的および発生段階特異的なmRNAの動態を調べた.胚発生における孵化準備期に2つのアイソザイムの発現が切り替わる現象を分子レベルで裏打ちすると同時に,本研究で得られたキイロショウジョウバエ,ヒト,ヤツメウナギのアルドラーゼに関する知見をも合わせて考案した.一方,特殊遺伝子として体液キモトリプシンインヒビターの遺伝子を解析し,多重遺伝子と多様なタンパク質成分との対応関係を明らかにした.ついで精巣特異的に発現されるDMC1と相互作用するタンパク質の遺伝子をsubtractionにより単離して解析し,高等真核生物としてははじめてATPase inhibitorタンパク質,ミトコンドリアATPase inhibitorタンパク,G-protein調節タンパク質などの遺伝子を単離した.さらに,九大保存の多様な突然変異系統,とくに発生異常系統について,微細構造およびタンパク質の動態を追求し多くの知見が得られたばかりでなく,クワコの染色体数における地理的ニ型を明確にした.以上,本研究で得られた成果は,ポストゲノム時代における分子遺伝学,発生遺伝学,進化遺伝学の対象としてのカイコの地位をより高めることに貢献するものと期待する.
