研究概要

1)キチン質製材の開発:水溶性キチンの作製を実施した。キチンは生体親和性が高いが、水には不溶であり、その溶媒は強酸である蟻酸しか報告されていない。この不溶性のキチンをカルボキシメチル化、硫酸化、リン酸化して水溶性のキチンを作製した(重政、斎本、指輪)。またリン酸キチンについては、低分子化したものも作製した。
2)水溶性キチンの獣医臨床への応用:これらの水溶性キチンをキトサンによる肺炎のブロッカーとして応用した。これらの水溶性キチンはすべてキトサン肺炎を程度の差はあるがブロックした。しかしその効果はリン酸キチンで最も強く認められた(南、岡本)。低分子化したリン酸キチンには肺炎の抑制効果をしましたが、高分子のものと比較して不十分な抑制効果であった。
3)水溶性キチンの肺炎ブロック効果のメカニズム:キトサンが多形核白血球の遊走因子でありかつ化学発光能を高めることを報告した。さらに、キトサンは補体の副経路を活性化し、産生されたC5aが重要な多形核白血球の刺激物質になることを証明した。リン酸キチンはキトサンの補体活性化能をブロックする機能はなく、肺炎防御のメカニズムは遊走能に対する直接的な効果であることを、証明した。水溶性キチンの一つであるリン酸化キチンは新しい抗炎症剤として期待のもてる物質であることを明らかにした。
