研究概要

Evi-1は2つのclusterに分かれzinc finger構造を持つ核内蛋白質をコードし、正常骨髄細胞ではほとんど発現が認められないが、白血病において高発現して造血細胞の悪性化に深く関与する。TGFβは、造血細胞を含めた種々の細胞の増殖を負に制御する代表的な増殖抑制因子である。PAI-1およびp15遺伝子はTGFβにより発現が誘導される遺伝子であるが、Evi-1はTGFβの細胞内シグナル伝達分子の一つであるSmad3と結合してTGFβによる両者のプロモーターの活性化を著明に抑制した。またTGFβは上皮細胞に対して著明な増殖抑制をかけるが、Evi-1を導入した上皮細胞はこのTGFβによる増殖抑制から逃れた。
以上のことからEvi-1はSmad3を特異的に不活化することによってTGFβシグナル伝達を抑制すると考えられた。さらに、我々はEvi-1がMAP kinaseの一つであるJNKと結合し、その活性を阻害することを明らかにした。JNKはUVや浸透圧などのストレス刺激やTNFなどにより活性化されるが、Evi-1はこれらの刺激によるJNKの活性化を抑制した。その抑制には第1zinc finger領域が必要であり、この領域を介してEvi-1はJNKと結合した。
これらのことから、Evi-1は活性化されたJNKと核内で結合し、JNKによる基質のリン酸化を阻害すると考えられた。UVなどの細胞外ストレス刺激によるJNKの持続的な活性化はアポトーシスの誘導に関与することが知られているが、Evi-1はUV照射によるアポトーシスの誘導を抑制した。このことから、Evi-1が細胞内において種々のストレスによるJNKの活性化を抑え、アポトーシスを抑制することによって悪性化に関与すると考えられた。
