研究概要

1, 我々が発見したRafのシステインリッチ領域とRasのアクチベータ領域との間の翻訳後修飾(ファルネシル化)依存性の第二の結合の認識機構と意義をさらに解析した。化学的にファルネシル化したRasのC末端対応合成ペプチドを用いて蛍光偏光度測定法により結合を解析し、Ras-Raf間の第二の結合結合に際してファルネシル基が認識されていることを証明した。さらに、Rasとがん抑制遺伝子産物Rap1A(Krev-1)によるRaf-1,B-Rafの異なった活性調節の分子機構の解明、および、Rap1AのプロテインキナーゼAによる燐酸化のRaf-1活性抑制作用に及ぼす影響の解析から、「Ras-Raf間の第二の結合の強度が適当であることがRafの活性化に必要」との仮説を証明した。
2, 出芽酵母アデニル酸シクラーゼについて、その結合蛋白質CAPのN末端36アミノ酸残基の領域とシクラーゼC末端領域の複合体がファルネシル化されたRasの第二の結合部位である事を遺伝学的、生化学的に証明し、結合認識機構を解析した。この結合がRasによるシクラーゼ活性化に必須である事を示した。この結果、エフェクター活性化においてRasの翻訳後修飾(ファルネシル化)が必須である分子機構が解明された。Rasの翻訳後修飾依存性の第二の結合の存在がエフェクターの種類によらず普遍的であることが示唆された。
3, 線虫C.elegansにおいて新規Rasエフェクター候補蛋白質PLC210を発見し、それがRas/Rap1A蛋白質とGTP依存性に結合する新種のホスホイノシチド特異的ホスホリパーゼCである事を証明した。PLC210の哺乳動物におけるホモログを発見してcDNAをクローニングし、そのコードする蛋白質が線虫PLC210と同じ活性を示すことを証明し、Ras/Rap1Aによる調節機構を解析した。
