研究概要	医薬品の多くは多環構造上に、様々な官能基が存在している。そこで効率的な多環構造構築法の開発は創薬研究において極めて重要な課題である。研究代表者は本目的に沿った分子内ダブルミカエル反応および分子内ミカエル・アルドール反応を開拓しており、これら反応の活用と展開を検討し、以下の研究成果を挙げることができた。 (1) クルモリンの全合成:シクロペンテノンの4位にα,β-不飽和エステル鎖を持つ化合物を、リチウムアミドで処理すると、2回の共役付加反応が連続的に起って、三環性の架橋化合物が立体選択的に生成した。さらに本成績体より3行程で目的とする天然物へと変換し、強力な抗カビ作用が報告されているクルモリンの超効率的な全合成を達成した。 (2) セドランジオールおよびセドラノキシドの全合成:シクロペンテノンの5位にα,β-不飽和エステル鎖を持つ化合物を分子内ダブルミカエル反応に付すことによって、三環性架橋化合物を高立体選択的に高収率で得ることができた。本成績体より、環拡大反応等を行って二種のセスキテルペノイドであるセドランジオールおよびセドラノキジドの全合成を完成した。 (3) タカモニンの合成研究:インドールの2位にα,β-不飽和エステル鎖、3位に共役不飽和アミド鎖を持つ化合物をアミンの存在下シリルトリフレートで処理することによって、インドロキノリジン体を一挙に合成した。本成績体は数行程でタカモニンの合成中間体へと導くことができ、ムスカリンM2レセプターに対して特異的に結合するタカモニンの短行程合成が可能となった。
