研究概要

(1)Crk癌遺伝子による発癌機構の研究
v-Crkによる細胞の癌化においては、PI3K/AKT経路の活性化が非常に重要な役割を果たしていることを明らかにした。さらにFocal Adhesion Kinaseのチロシンリン酸化とH-Rasの活性化が、このv-CrkによるPI3Kの活性化に必須であることを示し、v-CrkのSH2、SH3両ドメインが如何にして協調しながら細胞を癌化させるシグナルを生み出すのかを分子レベルで初めて明らかにすることができた。
(2)c-Crk IIがかかわる情報伝達経路の解析
c-CrkIIがRhoを活性化することを見出した。また、c-CrkIIが定量的にNIH3T3細胞をトランスフォームすることをはっきりと示し、トランスフォーミング活性がv-CrkだけにとどまらずCrkタンパクに共通の性質であることを明らかにした。
(3)ショウジョウバエを用いたCrk及びCrk結合因子の遺伝学的解析
ショウジョウバエにおいてCrkはthorax closureと呼ばれる発生過程で、PDGF/VEGFレセプターホモログからJNK pathwayへのシグナル伝達に関与していることを遺伝学的に初めて見出した。この過程にはCrkのSH3に直接結合するDock180と、その活性に必須な因子であるELMOが関与し、Racを介したJNK pathwayの活性化が起こることを明らかにした。
(4)非受容体型チロシンキナーゼc-Ablの制御機構の解析
c-Ablが、c-Srcやc-Crkなどのトランスに働くタンパクで活性の調節を受けることを示した。
また、c-Abl遺伝子を欠失した細胞は、正常な細胞よりもSV40T抗原の発現により癌化しやすいという現象を見つけ、c-Ablが癌抑制遺伝子として機能し得ることを見出した。
さらに、Abiがc-AblとMenaを結ぶアダプター蛋白として機能することを明らかにした。
(5)ヒト正常繊維芽細胞が示す癌遺伝子に対する抵抗性の解析
ヒト正常繊維芽細胞は、テロメラーゼの導入により不死化しても、なお、ラット正常繊維芽細胞に比べて活性化Ras+SV40T抗原の組み合わせによる癌化誘導に対して、より強い抵抗性を示すことを明らかにし、テロメラーゼの発現抑制以外の、ヒト細胞に特異的な、未知なる抗癌化機構が存在することが強く示唆された。
