研究概要

我々はC型肝炎ウイルスが肝細胞に与える影響を解明するために、C型肝炎ウイルスのコア遺伝子をマウスの肝細胞株BNL.CL2と同細胞由来のtransformed cellであるBNL.1MEA.7R.1に導入し、コア蛋白を発現するそれぞれ複数のパーマネントクローンを樹立した。これらのクローンの遺伝子発現プロファイルを1176遺伝子を搭載したアレイにて解析し、アレイ上3倍以上の発現の差を認めた遺伝子について、Northern blotにてその発現の差を確認した。これにより、コア遺伝子の発現によりCL2、A7R1の両細胞でともに発現が低下する遺伝子を2個、A7でのみ発現が上昇する遺伝子を3個同定した。これらの遺伝子はC型肝炎ウイルス感染の病態に密接に関連していることが推測されることから、これらの遺伝子の発現が肝細胞に与える機能的な意義について現在精力的に解析を行っている。また、これらのクローンを用いて、コア遺伝子の発現がCL2細胞、A7R1細胞の増殖とアポトーシスに与える影響を解析したところ、アポトーシスには影響を与えなかったが、両細胞で共に細胞増殖の遅延がみられ、これはG1期からS期への移行の障害によるものであった。これらの結果は、C型肝炎ウイルスの感染そのものは、肝細胞の増殖を阻害するものの、アポトーシスには影響を与えず、適切な細胞死シグナルを与えることによりC型肝炎ウイルスの除去が可能であることを示唆していると考えられた。
