研究概要

本研究はCpGメチル化を介したクロマチン構造の制御機構を、関与する蛋白質因子の立体構造と分子間相互作用の構造的基盤を基に解明することを目的とした。具体的には、研究対象として、メチル化DNA結合蛋白質であるMBD1、さらにヒトHR23B-プロテアソームS5aサブユニットユビキチン相互作用モチーフ(ubiquitin-interacting motif : UIM)複合体、ユビキチンのリジン48リンクの2量体、4量体、リジン63リンクの2量体、4量体、酵母Saccharomyces cerevisae Dsk2pのユビキチン会合ドメイン(ubiquitin-associated : UBA)-ユビキチン複合体を取り上げた。MBD1については、MBD1 M1行い、それからUIM-ユビキチン相互作用を推察した。また酵母Dsk2pユビキチン会合(UBA)ドメイン-ユビキチン複合体の構造解析を行った。加えて。Lys48リンク、Lys63リンクの2量体、4量体ユビキチン鎖の構造的特徴を調べるためにサブユニット特異的な安定同位体ラベルを行い、サブユニット間の相互作用面を化学シフト摂動法(chemical shift perturbation)、交差飽和法(cross saturation)を使って調べた。
