研究概要

本研究ではバイオミネラリゼーションを,結晶化が行われる場としてのマトリックス分子,結晶となる無機分子,そして結晶化の制御に関わる有機分子の3要素に分けて,これらの要素の相互関係を吟味することによりナノレベルでの結晶化制御の技術を開発することを目指した。強い炭酸カルシウム結晶化阻害活性をもつアカフジツボ・リンパ液レクチン(BRA-2)のN末端から39位アスパラギン残基に結合したハイマンノース型糖鎖を,グリコペプチダーゼFで除去し,BRA-2のカルシウム塩結晶化阻害活性,レクチン活性及びタンパク質分解酵素に対する安定性への糖鎖の役割を明らかにした。BRA-2とカルシウムイオンの相互作用におけるpH変化の影響のほかに,糖鎖の脱着によってもカルシウム塩結晶化の制御が可能であった。アカフジツボの殻形成に関わる有機マトリックス成分としてのレクチンの働きを解析した。真珠層を形成する貝殻のカルシウム塩結晶構造の特性を原子間力顕微鏡及びレーザー顕微鏡で精査し,アラレ石結晶構造の生物間個体差,殻成長点における多様性,光の反射率と結晶構造の相関を示した。
