研究概要

寄生現象において、自由生活型の祖先から出発し、寄生生活に移行してからの進化の過程おいて宿主内の環境に適応し、宿主特異性や臓器特異性をそなえた種々の寄生虫が成立したと考えられる。この様な観点から我々は回虫などの寄生虫と宿主であるヒトのミトコンドリアを用いて酸素適応機構の解明を目的として研究を進めている。回虫で見い出したNADH-フマル酸還元系は多くの寄生虫に存在し、宿主体内の環境で中心的な役割を果している事が明らかになった。この系は複合体I(NADH-ユビキノン還元酵素)、ロドキノンおよび複合体II(ロドキノールーフマル酸還元酵素:RQFR)の3成分から構成され、 NADHから最終電子受容体であるフマル酸への電子伝達を触媒している。その生理的意義は嫌気的グルコース分解系の最終ステップとして、無酸素下でも複合体1の共役部位を駆動する事によりATPを合成できる点にある。そこで本研究ではNADH-フマル酸還元系の分子構築とその生理機能の特徴を明らかにする目的で、回虫成虫ミトコンドリアを用い、研究を進めた。
1.新規の複合体1阻害剤を多数合成し、回虫酵素とウシ心筋酵素に対する阻害効果について徹底的な解析を加えた。その結果、回虫酵素はこれらの阻害剤に対する相互作用が宿主哺乳類と大きく異なる点を明らかにした。
2.複合体II(ロドキノールーフマル酸還元酵素)が生活環の中でサブユニット構成を大きく変化させる事を明らかにした。さらに成虫ミトコンドリアから酵素を精製し、結晶を得て解析した結果、ロドキノールからフマル酸への複合体内での電子伝達経路が明らかになった。また、複合体II特異的阻害剤アトベニンを見出した。
3.自由生活性線虫Caenorhabditis elegansの長寿命変異株clk-1のキノン組成の分析から合成中間体であるデメトキシユビキノンの蓄積を明らかにした。この事実をふまえ、clk-1の遺伝子産物がデメトキシユピキノンを水酸化するヒドロキシラーゼと考え、酵素活性測定系の確立をin vivoおよびin vitroで試みた。その結果、線虫clk-1が大腸菌の本酵素の変異株の増殖を回復させた点はCLK-1がヒドロキシラーゼである事を示している。
