研究概要

X線結晶構造解析法:機能性RNAとタンパク質との相互作用および共同作用のメカニズムを複合体のX線結晶構造解析によって明らかにした。遺伝暗号の翻訳においてtRNAとアミノ酸の正確な対応づけを担うアミノアシルtRNA合成酵素について結晶構造解析を行い、反応におけるtRNAの役割を明らかにした。アミノアシル化および誤って付加されたアミノ酸の校正いずれにおいてもtRNAは単なる基質ではなく、タンパク質に構造変化を誘起するなどして積極的に反応に参加していることが解明された。前駆体tRNAの成熟の構造基盤を明らかにするために、プロセシングおよび転写後修飾を担う酵素群について構造解析を行った。特にArcTGTとtRNAとの複合体から、tRNAは修飾の際にラムダ型と名付けた中間状態を経ることが明らかになった。また、生殖細胞の分化に関与するRNAヘリケースであるVasaとRNAとの複合体の構造解析に成功し、DEAD-boxRNAヘリケースがRNA二重鎖をほどく分子機構をはじめて明らかにした。
NMR法:ヒトおよび超好熱性古細菌のSRP RNAのhelix 6に由来するステム・ループの立体構造をそれぞれ決定し、SRP19との相互作用について考察した。高度好熱菌SmpBの溶液構造を決定し、さらに、tmRNAとの結合によりSmpBが構造変化することを示唆した。ウナギのLINE3'末端に存在するステム・ループの立体構造を決定し、GNRNAモチーフの構造およびステムの柔軟性が活性に重要であることを示唆した。また,RNA分子の立体構造をその特徴によって分類するシステムを開発し、NMR法による構造解析に応用した。
