研究概要

本研究の目的はピロリ菌(Hp)感染において、胃粘膜で発現するO-グリカンの役割を明らかにすることである。胃粘膜下層の腺粘液細胞から分泌される腺粘液は、非還元末端にα1, 4結合型N-アセチルグルコサミン残基を有するO-グリカン(αG1cNAc)を含んでいる。Hpは胃粘膜の表層粘液内に棲息し、腺粘液内には認められないことから、ピロリ菌に対するαG1cNAcの効果を解析した。αG1cNAcはHpの増殖や運動能を抑制すると共にHpに形態異常を引き起こすこと、Hpの細胞壁成分であるコレステリル-α-D-グルコピラノシド(CGL)はHpの生存に重要であることを示した。更に、CGLの生合成に関わる糖転移酵素(CHLαGcT)の遺伝子を発現クローニング法により同定、αG1cNAcがCHLαGcTの活性を阻害することを示し、αG1cNAcはCGLの生合成を抑えることでHpに対する抗菌的効果を示すことを明らかにした。αG1cNAcを欠損したα4GnT KOマウス、並びに胃粘膜表層にαG1cNAcを発現したα4GnT TGマウスの作出に成功した。
次に、Hpが感染した慢性活動性胃炎におけるリンパ球の浸潤メカニズムを解析した。この様な胃粘膜では、慢性炎の活動度が高度になる程、6-スルホシアリルルイスX含有O-グリカン(ssLeX)を発現した高内皮細静脈様血管(HEV様血管)の数も増加した。更にHEV様血管にはL-セレクチン-IgMキメラ蛋白が結合したことから、慢性活動性胃炎ではL-セレクチンを介したリンパ球ホーミングが生じていることが示された。
以上、胃粘膜で発現する2種類のO-グリカンであるαG1cNAcとssLeXはピロリ菌感染の病態形成において、それぞれ重要な役割を担っていることが明らかとなった。
