研究概要

コンビナトリアルケミストリーは、組み合わせの概念を用いることで、迅速な化合物ライブラリーを構築するための化学である。特に、その技術は、医薬品の開発研究および開発のスピードアップに大きな力を発揮している。現在では、コンビナトリアル合成によって、医薬品を含めさまざまな機能性分子開発のための化合物が合成されている。一方、天然から抽出される低分子有機化合物(いわゆる天然物)には、今なお新しい薬理活性を有するものが発見される。これは、天然物が有する大きな多様性が、人間の力による合成化合物群よりも遙かに大きいことを意味している。そこで、天然有機化合物をリードとするライブラリーを構築することができれば、その化合物群は、薬剤開発のリード化合物だけではなく、生物機能解明を行うためのケミカルブローブ開発に利用できると考えた。しかしながら、これらの天然物は、さまざまな官能基を有するため、現在の技術では、そのライブラリー構築は容易ではない。そこで、本研究では、固相および液相法を用いた複雑な官能基を有する天然物をリード化合物とライブラリー構築法の開発を行なった。その結果、25種の環状デプシペプチドオーリライド類縁体、74種類の抗腫瘍性海産性プロスタノイドクラブロン類縁体、122種類の16員環マクロラクトン化合物マクロスフェライドA類縁体、24種類のエルゲノシン298-A類縁体の固相ライブラリー構築を達成した。また、バンコマイシン類縁体の固相誘導体合成法の開発に成功した。さらに、液相ライブラリー合成にむけて、ワンポットグリコシル化反応を利用した大豆エリシター7糖、core2型糖アミノ酸、シアル酸含有糖鎖の合成法の開発に成功した。
