研究概要

本研究ではHIV-1のVifおよびNef、また、HIV-2のVpxおよびVprの機能領域について解析し、以下のような成果を挙げた。(1)HIV-1 Vif。エイズ長期未発症者と発症者由来のvif遺伝子の感染性付与機能を検討した結果、生体内でのウイルス複製とエイズの進行にVifの機能が関わることが示唆された。全領域にランダムに変異を導入した一連の変異体の解析中に、Vifが非常に速やかにプロテアーソム分解を受けることを見出し、これに強く関与するリジン残基を同定した。Vifのプロテアソーム分解高感受性はアクセサリー蛋白質中で特異的であるが、ウイルス感染性を至適に保つためには重要であることも確認された。(2)HIV-1 Nef。MHC-I分子のdown-regulation能のみを欠損した点変異体が感染したT細胞を用い、HIV-1 NefとHIV-1特異的CTL細胞との相互作用を解析した。NefはCTLの細胞傷害活性を効率的に抑制するが、サイトカイン産生は部分的にしか抑制しないことが明らかになった。(3)HIV-2 Vpx/Vpr。HIV-1ゲノムはVprをコードするが、HIV-2ゲノムはVprに加えてアミノ酸配列で相同性があるVpxもコードする。ホモロジーモデリングの結果、これらの蛋白質は3ないし4個のヘリックスからなる極めて類似した構造を持つ可能性があることがわかった。HIV-2のVpx/VprはHIV-1 Vprに比べ、ウイルスゲノム逆転写促進能やウイルス粒子放出促進能等多彩な機能を持つことを観察した。
