研究概要

本研究では、免疫遺伝学、構造生物学、比較ゲノム学、免疫生物学など幅広い視点から、ナチュラルキラー(NK)レセプターならびにNKレセプターと相互作用するMHCクラスI分子の構造・機能・進化を解析し、以下の成果を得た。
1.ヒトの主要なNKレセプターであるKIR (killer immunoglobulin-like receptor)遺伝子群はLRC (leukocyte receptor complex)領域に存在するが、第7染色体に位置するマウスのLRC領域にはKIRと相同な遺伝子群は存在しない。このため、マウスにはKIR相同遺伝子は存在しないというのが定説であった。われわれは、マウスのX染色体上にKIRと相同な遺伝子(Kirl : Kir-likeと命名)が2コピー存在することを明らかにした。
2.マウスLRC領域には、IgA Fc receptor遺伝子(Fcar)が欠損していることを明らかにするとともに、ラットとマウスではLRCのゲノム構造が大幅に異なっていることを示した。
3.げっ歯類以外の哺乳類では、KIRがNKレセプターとして使われているというのがこれまでの通説であったが、ウマではC型レクチン様分子であるLY49がNKレセプターとして使われていることを強く示唆する結果を得た。
4.無顎類ヌタウナギにおいて抗原レセプターと類似したVドメインを有するpaired Ig-like receptor遺伝子群(APAR)を同定した。本遺伝子群はNKレセプター様の機能を有している可能性が高い。
