研究概要

1.ヒトVα24陽性NKT細胞の体外増殖培養液を、10%FBS、10%リコンビナントヒト血清アルブミン、10%自己血漿、10%自己血清で比較したところ、10%自己血漿による培養が最も効率的であった。
2.全細胞数に占めるVα24陽性NKT細胞の割合を比較したところ、G-CSF投与前の末梢血では増殖比が約350倍であったのに比べ、G-CSF投与後の末梢血では増殖比が約1900倍であり、G-CSF投与による増殖能の改善が示された。
3.G-CSF投与前後の細胞と血漿をそれぞれ交叉組合せし培養したところ、「G-CSF投与後の細胞」に「G-CSF投与後の血漿」を添加した場合がVα24陽性NKT細胞の増殖比が最も高く、G-CSF投与後の細胞、および、血漿のいずれもがVα24陽性NKT細胞の増殖に関与していることが分かった。
4.G-CSF投与後末梢血由来のVα24陽性NKT細胞には、IL-3,IL-4,IL-7のレセプター発現が高まっていた。
5.G-CSF投与後の血漿中には、IL-3,IL-7,IL-13の産生が高まっていた。
6.培養後の細胞群には約20%のVα24陽性NKT細胞、CD4^+細胞、CD8^+細胞、次いで約10%のNK細胞が含まれ、B細胞、単球は殆ど含まれていないことが分かった。
7.G-CSF投与後の末梢血を用いて誘導したVα24陽性NKT細胞は、IFN-γ、および、IL-4の産生が高まっていた。また、Vα24陽性NKT細胞のサイトカイン産生は、IL-4よりもIFN-γ優位であった。
8.この培養系を用いて増殖したVα24陽性NKT細胞は、CD1d非拘束性に細胞障害性を示した。
