研究概要

本研究は、ライソゾーム酵素であるベータグルコシダーゼの先天的欠損によるゴーシェ病に対して、中枢神経系を標的とした低分子物質による新たな治療法を開発することを目的として行っている。昨年までに、ゴーシェ病の変異酵素のF213I変異を活性化するグルコース類似体N-octyl-valienamime(NOV)を見出した。本年度は新たにNOV類似の阻害剤13種類を合成し研究を進めた。F213I遺伝子異常をホモでもつ細胞に、これらの阻害剤を添加し4日間培養後にベータグルコシダーゼ活性を測定した。新しい阻害剤では、酵素活性を上昇させるものはなかった。さらに、NOVで活性が上昇する変異を新たにスクリーニングしN188S/G193Wを持つ患者細胞の活性が4倍に上昇することを見いだした。マウスモデルの開発として、アメリカジャクソン研究所から、ベータグルコシダーゼノックアウトマウスを購入し、ヒトベータグルコシダーゼcDNAにF213I変異を導入した後にpGCCIベクターに組み込み、F213I変異を持つモデルマウスの作成を試みた。F213Iを持つヒトcDNAの作成には成功した。しかしながら、ジャクソン研究所から購入したβグルコシダーゼノックアウトマウスのペアは正常であることが明らかになり、再度マウスの発注を行った。今後、動物モデルを確立し、治療研究を進め患者さんへ応用できるようにすることが重要と考えられた。
