研究概要

1.脂肪細胞分化と肥満発症におけるレニンアンジオテンシン系の生理的・病態生理的意義
高脂肪食負荷では標準食の場合と比較して、対照マウスの体重と脂肪組織重量と脂肪細胞肥大が認められたが、1型アンジオテンシン受容体欠損マウス(AT1a-KO)では明らかに抑制されていた。以上より、AT1a-KOマウスでは高脂肪食誘導性肥満症の発症が抑制されることが明らかになり、肥満症におけるAT1aの病態生理的意義が示唆された。
2.「脂肪組織リモデリング」の分子機構
独自に開発した脂肪細胞とマクロファージの共培養系を用いて、脂肪細胞に由来する飽和脂肪酸がマクロファージの炎症性変化を誘導し、これによりマクロファージにおけるTNFαの産生が増加して脂肪細胞における脂肪分解と炎症性変化を増大するという「悪循環」の存在を提唱した。更に、飽和脂肪酸は「TLR4の内因性リガンド」としてマクロファージに炎症性変化を誘導すること、TLR4シグナルの阻害により肥満の脂肪組織における炎症性変化と糖代謝障害が改善することを証明した。一方、代表的なn-3多価不飽和脂肪酸であるEPAは飽和脂肪酸により誘導される炎症性変化に強力に拮抗すること、メタボリックシンドローム患者において高感度CRPや動脈硬化惹起性リポタンパクを減少すること、脂肪組織の炎症性変化を抑制することにより血中アディポネクチン濃度を増加することを見出した。又、脂肪細胞の肥大化によりMCP-1やIL-6の発現が亢進し、アディポネクチンの発現が減少することを明らかにし、脂肪細胞の肥大化に伴うMCP-1の発現亢進にはMAPKの負の制御因子であるMKP-1の発現低下によるERKの活性化が関与することを見出し、肥大化した脂肪細胞にMKP-1遺伝子導入することにより、MKP-1の増加とともにERKの活性とMCP-1産生が減少することを証明した。
