研究概要

有顎脊椎動物に限定される獲得免疫とちがって、未だに起源の明らかでない自然免疫の進化を解明する目的で、補体系の進化的な起源を解析した。補体系の中心成分であるC3を含むチオエステル含有タンパク遺伝子に汎用できる縮退プライマーを設計し、数種類の前口動物、刺胞動物を解析し、イソギンチャクからはC3とα2Mの両者を、ヒドラ、カタツムリ、ホシムシからはα2Mを同定した。この結果はC3とα2Mの起源及びお互いへの分化は刺胞動物の出現以前にまで遡れること、及びα2Mは多くの動物群に保持されているが、C3は進化の過程で何度も失われたことを示している。ホヤ補体系の解析により、α鎖、β鎖よりなるインテグリン型の補体受容体の存在を明らかにし、それがC3と結合することにより異物の貧食に働いていることを証明した。また、ホヤ補体遺伝子のゲノム構造の解析では、C3とBf遺伝子が連鎖していないことを明らかにし、脊椎動物MHCにみられるクラスIII補体領域は脊椎動物の系統内で形成された可能性が高いことを示唆した。さらに、数種類のサメを用いた解析により、C3,C4,C5遺伝子が存在すること、ただしC5は種によっては擬遺伝子化していることを明らかにし、これら遺伝子間の遺伝子重複を軟骨魚類の出現以前に位置付けた。以上の結果より、補体系の中枢部分は10億年以上前に確立され、その後、後口動物の系統でほとんどの成分がそろい、さらに有顎脊椎動物の出現に際して更なる遺伝子重複により今日哺乳類に見られるような高度に組織化された系に進化したことが示唆された。
