大腸菌RNaseGの機能解析とmRNA安定性制御によるタンパク質発現系の構築

研究概要

大腸菌のエンド型リボヌクレアーゼRNase GによるmRNAの安定性制御のメカニズムを明らかにするために、RNase GのadhE mRNAに対する作用を調べた。その結果、-17部位でRNase Gによる切断を受けたmRNAは速やかに分解され、-31部位でRNase IIIによって切断を受けたmRNAは翻訳されることが示唆された。また、RNase G変異株では解糖系酵素のmRNAの安定化によりピルビン酸の過剰合成が起きていることを見いだした。この知見をもとに、RNase G欠損株を用いたピルビン酸及びバリンの生産法を開発した。エンド型リボヌクレアーゼRNase Eは大腸菌の生育に必至であるが、細胞分裂タンパク質FtsZをコードするmRNAのプロセシング異常がRNase E欠損の致死性の原因であることを明らかにした。RNase Eによる切断を受けないftsZ mRNA前駆体からの翻訳はRNA結合タンパク質Hfqにより阻害されていた。このようなRNA代謝の制御により遺伝子発現が調節を受けているものを探索した結果、酸耐性に関わる遺伝子群や酸化ストレス防御に関わる遺伝子を見いだした。RNase Eの温度感受性変異株rne-1やhfq変異株は酸や酸化ストレスに対する感受性が増大していた。これらのことから、細菌細胞ではこのようなRNA代謝の制御による遺伝子発現調節がかなり広汎に行われていることが推測された。
