研究概要

15年度の研究においては、ゲノミックプローブによるカイコ染色体同定とヘテロクロマチンによる性染色体の簡便な同定法ならびに雌特異的プライマーから特定されたBAC(W-BAC)を用いて鱗翅目昆虫において最初となるBAC-FISH法の開発に成功した。この成果を応用して、17年度研究ではGISHとテロメアFISHを組み合わせたマルチカラーFISHによる性染色体構成の簡便な判別法を開発するに至った。カイコBAC-FISH法をさらに改善すると同時に、カイコRAPD連関地図へと座位決定を進めたBACクローンを使用して、連関地図に対応したカイコ染色体の個別同定を可能とした。さらには、16年度研究において全染色体一括認識と核型検定(カリオタイピング)を行えるシステムの構築を行い、80年以上の長きにわたり不可能であったカイコ核型を明らかにした。平成17年度までに連関地図の充実化を進め、合計512個からなるBACコンティグから成る次世代に通用する分子連関地図が完成した。この地図は、突然変異形質による従来の連関地図と対応付けをほぼ完了しているとともに、データベース上に公開された既知遺伝子の大部分について対応するBACを検出したことから、突然変異の原因となる遺伝子のクローニングなどに活用が見込まれる。
17年度を中心にBAC-FISHによる染色体マッピングの拡充を行った結果、使用したBACのほとんどは組み換え価に基づいたRAPD地図上に座位決定された結果と良く一致したものの、連関地図上とは異なる染色体上にシグナルの認められるものや同一染色体上にマッピングされるが、位置関係に齟齬のあるものが検出された。これらは座位決定の誤りの可能性があるために対応連関群ならびに組み換え価の再検討が今後必要であると考えられる。
