研究概要

熱帯地域で流行を繰り返し多くの人的被害が発生しているデング出血熱重症化に関与する宿主および病原体因子を解明することを目的として、東南アジアのデングウイルス活動地域において、患者血清からウイルス分離しその遺伝子、生物学的性状を解析する一方、デング感染者のウイルス感染細胞の分析を実施した。フィリピンでは2000年前後にそれまで流行していたデング2型ウイルスが他の地域から侵入したデング2型ウイルスに置換する現象が観察され、ベトナムやバングラデシュにおいては地域に古くから生息しているデングウイルスが変化(進化)をしながら流行を繰り返していた。デングウイルスは遺伝子的には比較的急速な変異を繰り返しつつ地理的にも移動して時には勢力を拡大しているという生態学的特長が明らかにされた。今回の調査ではバングラデシュ、フィリピンで重症者から分離したウイルスと軽症者から分離したウイルスでは重要な遺伝子配列の差異は発見できなかった。しかし同一患者から異なる生物学的特徴(感染細胞嗜好性や増殖度)を有する2種のウイルス株が分離された、この発見はデングウイルスは複合集団として存在しているのであり、その病原性の解析にはウイルスと宿主固体の組み合わせ、およびウイルス集団のそれぞれのグループの増減等を考慮して解析する必要性が示唆された。したがって、デングウイルスと病原性の関連性の解析は従来考えられていたよりもさらに複雑な因子(一患者中のウイルス遺伝子の多様性)を考慮する必要性をしめしている。今後、アジアにおけるデングウイルスの動向を継続的に観察するとともに、さらに網羅的なデングウイルス関連蛋白質の生理作用解析がデング出血熱発生のメカニズムを解明するうえで必要である。
