研究成果の概要

本研究では、SLEにおけるるIRF7とIRF8の役割をプリスタン誘発性SLEモデルマウスを用いて検討した。IRF7欠損マウスは野生型マウスと同程度に糸球体腎炎を発症したが、自己抗体の産生は認められなかった。IRF8欠損マウスでは、自己抗体の産生はみられず糸球体腎炎も軽減した。野生型、IRF7欠損マウスにおいては腹腔内、腎臓等に炎症性単球が浸潤しており、炎症性サイトカインを産生していたが、IRF8欠損マウスでは浸潤がみられなかった。自己抗体の産生にはI型IFN経路が、糸球体腎炎等の臓器障害には炎症性単球由来のNF-κB経路依存性の炎症性サイトカインが重要な役割を果たしていることが考えられた。
