研究成果の概要

魚類性決定において、水温がどのように遺伝的性を上書きするのか、その分子機構を明らかにすることを目的とした。通常の雌(XX)と低水温により性転換を誘起した雌(XY)由来の生殖腺を用いてRNA-seqを行い、両区で有意に差がある９９遺伝子を単離した。IPA解析を行った結果、両者に有意な差があると認められたパスウェイは、calcium-signaling、protein kinase A signaling、opioid signalingであり、estrogenとTGF-β1がこれらの上流制御因子であることが示唆された。これら因子は低水温による遺伝的雄の雌への性転換機構に関与していると考えられる。

研究成果の学術的意義や社会的意義

本課題により、低水温により魚類の遺伝的雄（XY）を雌（卵巣）に性転換させるには、estrogenおよびTGF-βシグナル系が深く関わっていることが明らかとなった。本成果は、魚類のみならず、爬虫類等の他の脊椎動物における温度依存型性決定機構の全容解明、とりわけ低水温が誘起する遺伝的雄の性転換機構の解明に寄与すると期待される。また、魚類を含む脊椎動物の性決定機構の進化を解読するうえでも重要な分子基盤になると期待される。
