C型肝炎ウイルスの感染初期過程の分子機構

研究概要

HCVの受容体候補分子であるhCD81を全く発現していないHepG2細胞にHCVのエンベロープ蛋白質を持ったシュードタイプVSV (HCVpv)が感受性を示すことから、HCVの感染にhCD81以外の細胞表面因子の関与が示唆された。HCVpvの感染はヒト繊維芽細胞成長因子(hFGF)、特にFGF2とFGF7によって阻害され、可溶化型のhFGF受容体5(hFGFR5)がHCVpvの感染性を特異的に阻害した。また、hFGFR5を恒常的に発現するCHO細胞株はHCVpvの感染を許容できるようになり、siRNAによってhFGFR5をHepG2細胞からノックダウンさせると感受性の低下が観察されたことから、hFGFR5がHCVpvの新規受容体であることが示唆された。これまでは、CHO細胞で発現させたHCVエンベロープ蛋白質を被ったシュードタイプウイルスを用いて解析を進めてきたが、293T細胞で作製したシュードタイプレトロウイルスはhCD81依存的な感染性を示し、HepG2細胞には全く感受性を示さなかった。そこで、HCVのエンベロープ蛋白質を発現させたCHO細胞と293T細胞を用いて、それぞれHCVpv/CHOとHCVpv/293Tを作製した。両シュードタイプウイルスとも高マンノース型のエンベロープ蛋白質を保持していた。HCVpv/CHOはhFGFR5依存的にHepG2細胞に高い感染性を示したのに対し、HCVpv/293TはhCD81依存的にHuh7細胞に高い感染性を示した。また、慢性C型肝炎患者血清中にはHCVpv/293Tに対する高い中和抗体が高率に検出されたのに対し、HCVpv/CHOに対する中和抗体価は低かった。患者血清中に存在する天然のHCV粒子も、HCVpvと同様にHepG2細胞とHuh7細胞に結合した。HCV粒子のHepG2細胞への結合は可溶型hFGFR5や抗hFGFR5抗体で、また、Huh7細胞への結合は抗hCD81抗体によって阻害された。
