汎用性抗原提示人工細胞とペプチドによるヒト細胞障害性T細胞簡易誘導法の開発と応用

研究概要

日本人に対し汎用性の高い抗原提示細胞として、HLAA24保有者のEBV不死化B細胞株(B-LCL)を使用し、伝染性単核症、慢性活動性EBV感染症をモデルとして、EBV特異的な細胞障害性T細胞(CTL)の確認、試験管内増幅、および細胞障害活性について検討した。
1.EBV特異的なCTLの確認
用いたEBV特異的なテトラマーの中では、健常人、伝染性単核症患者の末梢血にBRLF1テトラマー陽性のCTLの存在を確認した。慢性活動性EBV感染症(CAEBV)患者末梢血からもBRLF1テトラマー陽性のCTLの存在が示された。
2.HLAA24陽性者由来抗原提示細胞刺激による抗原特異的CTLの試験管内誘導
HLAA24特異的抗原提示細胞に対しBRLF1ペプチド刺激の有無により、BRLF1特異的なCTLの誘導を試みた。ペプチド刺激の有無に関わらずBRLF1特異的CTLは誘導された。これは、抗原提示細胞から十分な刺激が、ペプチドの有無に関わらずCD8+CTLに入るためと考えられた。このことは、健康者およびCAEBV患者の両方で確認された。また、CD8陽性T細胞の試験管内増殖は、ペプチドパルスの有無に関わらず認められた。
3.誘導された抗原特異的なCTLの細胞障害活性の検討
ペプチド刺激の有無に関わらず、HLAA24陽性健常者から検出されるBRLF1テトラマー陽性GTLは、BRLF1ペプチド抗原で刺激されたときにのみ、CD8陽性CTLにインターフェロンγ(Inf-γ)産生細胞を誘導する。一方CAEBV患者では、健常者と同様にBRLF1テトラマー陽性CTLが誘導されて来るが、テトラマー陽性CD8陽性CTLのBRLF1ペプチド刺激によるInf-γ産生細胞は、健常者の約50%に比し、約30%と低下していた。
今回の研究により、CAEBV患者では健常者と同等の時間経過でBRLF1特異的なCTLが誘導されてくるが、そのCTLは、健常者に比較しInf-γ産生能が低下している可能性が示唆された。この機序の解明は、CAEBVの発症機序とCTLによる治療を考える上で重要な知見となろう。
