研究概要

われわれは、先に枯草菌のL31とそのパラログであるYtiAが、亜鉛濃度により巧妙に制御されていることを明らかにした。つまり、亜鉛が十分培地中に存在する時には、亜鉛を結合したL31のみが50Sサブユニットに結合し、yliA遺伝子はZurレプレッサーにより転写が抑制されているが、亜鉛欠乏下には、ZurレプレッサーはytiA遺伝子のオペレーターに結合することができずに、ytiA遺伝子の転写が誘導され、新たに合成されたYtiAタンパク質がL31を結合した50SサブユニットからL31を追い出しYtiA結合型のリボソームが機能することになる。一方、50Sサブユニットから追い出されたL31は不安定となり分解されて亜鉛を細胞内に供給することになる。この機構は、亜鉛欠乏時の緊急避難的な細胞内亜鉛濃度維持機構のひとつとして機能しているものと推察される。
いまひとつの亜鉛結合型リボソームタンパク質S14(rpsM)と、そのパラログである亜鉛非結合型リボソームタンパク質YhzAは、L31とYtiAと同様に亜鉛濃度によりZurレプレッサーを介してそれらの遺伝子の発現が調節を受けていることを明らかにした。L31-YtiAの場合と異なり、S14は30Sサブユニットのアッセンブリーに重要な役割を担い、生育に必須のタンパク質である。事実、S14の欠失した細胞では30Sと50Sサブユニットの異常な蓄積が見られた。蓄積した30SサブユニットにはS2とS3タンパク質がほとんど認められなかった。このことは、試験管内で行われたアッセンブリーマップと一致した結果であることを示している。さらに、S14の機能は亜鉛非結合型のYhzAタンパク質を強制発現させることにより効率は悪いが相補できることを明らかにした。
