研究概要

インテグリンが細胞増殖因子受容体との相互作用は、細胞の増殖・移動・生存などの生理的な過程のみならず、癌細胞の増殖・転移・浸潤など病理的な過程に重要な役割を担っている。一方、糖鎖は多くの膜蛋白質を修飾し、その機能を制御することが明らかになっているが、細胞膜上での分子間の複合体形成における役割に関してはほとんど不明である。その分子的基盤の解明は生理的な細胞内情報伝達システムや、またそのシステム破綻による癌化などの原因解明に不可欠である。本研究の目的は、インテグリンのアスパラギン型糖鎖に焦点を絞り、改変される糖鎖構造を持つインテグリンが細胞の増殖・接着・運動への影響と、それに共役した増殖因子受容体を介するシグナル伝達の変化の解析を通じて、インテグリン-増殖因子受容体の複合体形成に重要な糖鎖上のモジュールを明らかにすることにある。我々はこの2年間の研究を通じて以下の糖鎖機能が明らかとなった。1)糖転移酵素GnT-IIIを導入した癌細胞株では、細胞接着や細胞浸潤能が抑制されることが明らかとなった。それの結果はbisecting GlcNAc糖鎖がインテグリンに抑制的に働くことを示唆した。2)インテグリンα5サブユニットの機能制御に関わる糖鎖付加部位を特定した。3)インテグリンα3サブユニットの機能制御にコアフコースが必要である。Fut8欠損細胞においては、インテグリンα3の機能が低下していたが、Fut8の再導入でα3の機能が回復された。4)コアフコース糖鎖付加は増殖因子受容体EGFRの機能発揮およびインテグリンとの複合体の形成にも重要であることを明らかにした。
