研究概要

主にアフリカツメガエル(以下、ゼノパス)をモデル生物として用い、受精の分子機構、特に受精に伴い活性化するSrc型チロシンキナーゼxykの分子実体、およびxykの上流因子、すなわち精子が卵xykを活性化する機構、の2点に重点を置いた解析を行った。その結果、以下の成果を得た。受精卵において、細胞膜ラフトに局在する新規のチロシンリン酸化タンパク質uroplakin III(以下、UPIII)を同定した。UPIIIはアミノ酸残基数265の単一膜貫通タンパク質で、アミノ末端側から細胞外ドメイン(191アミノ酸)、膜貫通領域(28アミノ酸)、そして細胞内ドメイン(46アミノ酸)からなる。チロシンリン酸化は細胞内ドメインのTyr-249に起こることがMS/MS質量分析から明らかとなった。また、この分子が卵表面に存在し受精における精子との相互作用に関与することが明らかとなった。UPIIIのチロシンリン酸化は受精により活性化するゼノパスSrcとUPIIIを共発現させたHEK293細胞において再構成できることも明かとなった(J Biol Chem 2005)。さらに、精子と卵UPIIIの相互作用には、精子由来のプロテアーゼによるUPIII細胞外ドメインの切断という事象が関わっていることも示された。この切断反応を阻害する合成ペプチドが、受精そのものを阻止することも明らかとなった(Dev Biol 2005)。UPIIIはこのように、細胞外ドメインが精子との相互作用に、細胞内ドメインが受精に伴い活性化するSrcチロシンキナーゼのシグナル伝達に、それぞれ関与することが示唆された。xykのマススペクトロメトリによる分子構造の解析と系統発生学的な解析に関する論文を発表し、xykがゼノパスSrc遺伝子産物であることを明らかにした(以降、xykをxSrcと改称)(J.Biochem.2006)。卵細胞膜マイクロドメインに存在する精子プロテアーゼ受容体UPIIIを中心とするタンパク質複合体の機能解析を行ない、テトラスパニン型結合パートナーであるUPIbとの相互作用による細胞膜マイクロドメインへの局在、ガングリオシドGM1との特異的な相互作用、受精に伴い活性化するチロシンキナーゼxSrcに対する負の活性制御機構への関与を明らかにした(Genes Cells 2007)。
