研究概要

本研究課題の癌腫および組織型特異的な放射線感受性増感の分子標的探索について、我々は前立腺癌について以下の結果を得ている。前立腺癌はそのホルモン感受性のため内分泌療法が腫瘍縮小に有効であり、放射線治療の併用で幅広い病期で治療成績向上が得られている。しかし、ホルモン感受性または依存性腫瘍においてホルモンの存在が放射線に対する細胞応答に影響することは推測できるが、必ずしも明確とはなっていない。そこでホルモン感受性ヒト前立腺癌細胞株(LNCaP:野生型p53)においてアンドロゲン刺激(dihydrotestosterone: 以下DHT)やその阻害(Hsp90シャペロンコンプレックス阻害剤radicicol: 以下RD)が放射線感受性にどのように影響するかを検討した。
DHT単独の増殖能への影響については、DHT0.1nMまたは1nMの接触で有意な増殖能亢進が認められ、放射線感受性はDHT1nM存在下で照射すると低下した。RD500nMをDHTに併用するとDHTによる増殖能充進は抑制され、RDをDHTに併用すると、DHTによる効果が阻害され,相乗的な放射線増感作用が認められた。
上記の機序をアンドロゲン受容体(AR)、前立腺特異抗原(PSA)の発現の変化から検討すると、ARおよびPSA共にRD単独またはDHTとRDの併用で蛋白レベルの発現低下が認められた。Real-time定量性PCRではDHTまたはRD単独,DHT+RD併用のいずれでもmRNA発現レベルに有意な変化は認められず、免疫沈降法でRDの存在下でのHsp90とARの結合抑制が認められた。照射単独ではHsp90とARの結合には影響はなかった。そのため、上記の結果はmRNA発現抑制による蛋白合成抑制ではなくHsp90シャペロンコンプレックス阻害に伴うAR分解亢進によるものと考えられた。放射線の併用でも同様にDHT+RD併用でAR, PSAの発現低下が認められた。これらの結果よりHsp90シャペロンコンプレックスはホルモン感受性前立腺癌の放射線応答に重要な役割を果たしており、放射線感受性増感の分子標的となりうる可能性が示唆された。
