研究概要

カタユウレイボヤを主な材料として、精子活性化・誘引物質(SAAF)がもたらす精子運動活性化と走化性反応の分子機構を調べるとともに、ヒト・マウス等の哺乳類精子が卵及び精漿由来成分にどの程度に運動調節を受けるか、基礎的な研究を行っている。
1)SAAF受容体の同定:昨年に引き続きSAAF受容体の精製を行った。SAAFアフィニティカラムを作成し、精子細胞膜画分のタンパク質より精製を試みたところ、約300kDaのタンパク質がSAAFと相互作用することが判った。現在アミノ酸配列の同定を行っている。
2)SAAFによる精子鞭毛運動調節機構の解析:運動中精子のイメージングを行う蛍光顕微鏡装置の開発を行い、LEDストロボ照明を用いて毎秒50コマ、1コマ1ミリ秒の速度でイメージングを行うことを可能とした。そして卵より精子誘引物質が精製、同定されているカタユウレイボヤにおいて、走化性時の精子鞭毛運動について詳細な運動解析と細胞内カルシウムイメージングを行い、鞭毛内のカルシウム濃度の一過的な上昇が鞭毛打の対称性を調節し走化性行動を引き起こすことを発見した。
3)哺乳類における精子鞭毛運動調節機構の解析:マウスを用いて、精子運動に対する精嚢腺由来物質の阻害効果を研究している。昨年度我々が同定したマウス精子受精能破壊因子セメノクロチンによる精子の受精能阻害機構を調べるため、本年度はセメノクロチンの精子上の受容体を探索した。その結果、セメノクロチン受容体は脂質構成成分であるガングリオシドGMIであることを明らかとした。
