研究成果の概要

ユビキチンは構造安定性が高く可溶性も高いタンパク質であるが、ポリマー化したポリユビキチン鎖は熱や流体力学的応力により、アルツハイマー病をはじめとする神経変性疾患に関与する線維状凝集体を形成する。本研究はポリユビキチン線維の構造と形成機序を核磁気共鳴法により原子分解能で解析した。線維反応中間体の作製や重水素交換実験による表面構造解析を通じて、線維形成において重要な構造変化や分子間相互作用部位を特定した。また、新規レオロジー核磁気共鳴法を開発し、線維形成下におけるリアルタイム構造解析を行なった。

研究成果の学術的意義や社会的意義

長年多くの病理学者たちが抱いてきた不思議は、神経変性疾患で観察される異常タンパク質凝集体（封入体）のほとんどにユビキチンが含まれていることである。本研究は、どのようにしてポリユビキチン鎖が線維状凝集体を形成するかを原子分解能で解析したものであり、得られた研究成果は封入体の形成機構の理解、つまり神経変性疾患の発症機構の理解の一助となると考える。また、得られた構造学的情報は、熱や流体力学的応力によるマルチドメインタンパク質の構造変化や分子間相互作用を理解する上で重要な知見となり得ると考える。
