強皮症皮膚線維芽細胞におけるコラーゲン遺伝子転写制御・情報伝達の研究

研究概要

強皮症培養線維芽細胞ではコラーゲンなどの細胞外マトリックス発現亢進によって線維化していると考えられているが、その原因としてTGF-βが主役となっていると考えられ、その機序を明らかにする事は非常に重要である。TGF-β情報伝達のnegative feed back機構を司る癌遺伝子SnoNについて検討した。正常および強皮症培養線維芽細胞を用いて免疫ブロット法にてSnoN蛋白の発現量を検討したところ強皮症線維芽細胞において発現が亢進していた。Northern blot法にて強皮症線維芽細胞においてSnoN mRNAの発現が亢進していた。皮膚組織を用いた免疫染色法にて強皮症線維芽細胞におけるSnoNの発現亢進がin vivoで確認された。また正常線維芽細胞ではTGF-β刺激にてSnoNの発現が亢進したが、強皮症線維芽細胞ではSnoNの発現が恒常的に亢進しており、TGF-β刺激にてSnoNの発現に変化はなかった。さらに正常線維芽細胞ではTGF-β刺激にてSnoNとSmad3およびSkiとSmad4の介合が亢進したが、強皮症線維芽細胞では恒常的にSnoNとSmad3およびSkiとSmad4の介合が亢進しており、TGF-β刺激にて変化はなかった。また正常線維芽細胞ではTGF-β刺激にて亢進したI型コラーゲン蛋白発現、I型コラーゲンmRNA発現、a2(1)コラーゲン遺伝子転写活性が、SnoNの強発現によって全て抑制された。しかしながら、強皮症線維芽細胞では恒常的に亢進したI型コラーゲン蛋白発現、I型コラーゲンmRNA発現、a2(1)コラーゲン遣伝子転写活性が、SnoNの強発現によって全く抑制されなかった。以上の結果は、強皮症線維芽細胞におけるTGF-β情報伝達の異常を示し、強皮症培養線維芽細胞ではautocrine TGF-βsignaling(強皮症線維芽細胞自身が分泌るTGF-βが活性化してTGF-β情報伝達が亢進した結果)によってコラーゲンなどの細胞外マトリックス発現亢進によって線維化していると考えられているが、SnoNの過剰発現はautocrine TGF-βsignalingによるものであり、さらにautocrine TGF-βsignalingの維持に関与するものと考えられた。
