Aims of the proposed research are to observe activation of some genes related to Induced Resistance Systemic (ISR) or Jasmonate pathway, additionally to systemic acquired resistance (SAR) in Heat shock-induced resistance (HSIR), and to study whether heat shock proteins play roles in HSIR and the time if those HSPs take involve in HSIR mechanism. Also, to study whether the expressed genes possess particular heat shock element (HSE) that possibly involve in heat shock transcriptional factors (HSF). As the result of this study, we investigated HSIR against cucumber powdery mildew and confirmed initial mechanism of HSIR prior to SAR. Gene expression of Hsp70 were upregulated at 12 hour, followed by peroxidase and lipoxygenase at 24 hour after heat shock treatment. Putative HSE motifs were found in upstream area of those target genes. HSFs such as HsfA2, HsfB1 and HsfB4 gene expressions increased from 6 h after heat shock, which confirm that upon heat stress, some Hsfs were activated to bind HSE and upregulate transcription of Hsp70 and defense-related genes. From the results of previous study, HSIR has been considered to include SAR. We found a time lag between the expression of marker genes concerning these two pathways that suggested they were independent reactions that influence each other.
